Carrier Mediated Transport of Amino Acids, Their Drug Analogs*

چکیده
The oral absorption of amino acids, small peptides and their analogs has been studied in rats using single pass intestinal perfusion, The experimental data was analyzed using a modified boundary layer method permitting the calculation of intrinsic membrane absorption parameters. Cefatrizine, L-Dopa, cu-methyldopa, and L-PHE demonstrated concentration dependent absorption. Furthermore, the absorption of the amino acid analog, L-Dopa, was significantly inhibited by L-Leucine. Cefatrizine absorption was significantly inhibited by the peptide PHE-PHE (20 mM) whereas the amino acid L-PHE (100 mM) did not inhibit its absorption. The results clearly demonstrate that L-Dopa and a-methyldopa oral absorption occurs via the amino acidpathway, while cefatrizine absorption occurs via the peptide pathway. These results indicate that nutrient pathways may function as a possible route for the oral delivery of selected drugs.

Sodium-dependent neutral amino acid transport by human liver plasma membrane vesicles.
The activities of several selected Na(+)-dependent amino acid transporters were identified in human liver plasma membrane vesicles by testing for Na(+)-dependent uptake of several naturally occurring neutral amino acids or their analogs. Alanine, 2-(methylamino)isobutyric acid, and 2-aminoisobutyric acid were shown to be almost exclusively transported by the same carrier, system A. Kinetic anal...
متن کاملA Kinetic Investigation of a Carrier-Mediated Transport through a Bulk Liquid Membrane
The kinetics of the potassium thiocyanate transport mediated by dicyclohexyl-18-crown-6 (L) through a bulk liquid membrane is studied experimentally and theoretically. The proposed model is based on the assumption of a pure diffusion of the complex salt [K·L]+SCN¯ through the liquid membrane stagnant films at the interfaces. It illustrates the ...
متن کاملEstimation of apparent L-amino acid diffusion in porcine jejunal enterocyte brush border membrane vesicles.
There is an overlap of carrier-mediated L-amino acid transport and apparent simple diffusion when measured in intestinal brush border membrane vesicles. Using L-threonine and L-glutamine as representative amino acids, this study was undertaken to estimate apparent simple diffusion of L-amino acids and to establish the effective dosage of HgCl2 for completely blocking carrier-mediated L-amino ac...
متن کاملMechanism and Regulation of Isoleucine Excretion in Corynebacterium glutamicum.
Whole cells of Corynebacterium glutamicum were loaded with high cytoplasmic l-isoleucine concentrations, and isoleucine excretion from these cells was studied in terms of mechanism and regulation. The transmembrane isoleucine flux could be differentiated into carrier-mediated uptake, carrier-mediated excretion, and diffusion. After discrimination from the other transmembrane solute movements, t...
متن کاملEvidence for active transport of melphalan by two amino acid carriers in L5178Y lymphoblasts in vitro.
Investigation of the mechanism of uptake of melphalan by L5178Y lymphoblasts has been extended with particular emphasis on the chemical specificity of the transport mech anism. Evidence was obtained that uptake of melphalan was an active carrier-mediated process. Uptake appeared to proceed “uphill― against a concentration gradient of approximately eight-fold and was temperature sensitive, p...
